Influence of Hyaluronic Acids of Different Molecular Weights and Concentrations on the Enzymatic Activities of the Glucose Oxidase-mediated Peroxidase System

Abstract
Hyaluronic acid has been used as a constituent of oral health care products. The purpose of this study was to investigate
influences of hyaluronic acids of different molecular weights and concentrations on the enzymatic activities of the glucose
oxidase-mediated peroxidase system. Hyaluronic acids of four different molecular weights (10 kDa, 100 kDa, 1 MDa, and 2
MDa) and reagents containing the glucose oxidase-mediated peroxidase system were used for assays. The enzymatic activity of the glucose oxidase-mediated peroxidase system was examined by measuring oxidized o-dianisidine production. The four kinds of hyaluronic acids at 0.5 mg/mL did not affect the enzymatic activities of the glucose oxidase-mediated peroxidase system. Among the three kinds of specific molecular weight-concentation conditions (100 kDa-5.0 mg/mL, 1 MDa-0.5 mg/ mL, and 2 MDa-0.2 mg/mL) showing viscosity values similar to human whole saliva, only the 100 kDa-5.0 mg/mL condition inhibited the enzymatic activities of the glucose oxidase-mediated peroxidase system significantly (P < 0.05). Mixtures of hyaluronic acids with different molecular weights did not affect the enzymatic activities of the glucose oxidase-mediated peroxidase system. In conclusion, hyaluronic acids and the glucose oxidase-mediated peroxidase system could be used together in oral health care products. However, possible interactions between the molecules need to be considered.
